Abstract
Staphylococcus epidermidis is a normal skin flora bacterium that can act as an opportunistic pathogen and cause various infections, particularly those associated with the use of medical devices. The increasing incidence of bacterial resistance to antibiotics has encouraged the exploration of natural antibacterial agents. Broccoli (Brassica oleracea var. italica) contains several bioactive compounds, including flavonoids, alkaloids, saponins, and tannins, which are known to possess antibacterial properties. This study aimed to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of broccoli ethanol extract against Staphylococcus epidermidis. This study was conducted using a laboratory experimental design. Broccoli was extracted by maceration using 96% ethanol as the solvent. Phytochemical screening was performed to identify the presence of secondary metabolites. The antibacterial activity was evaluated using the broth dilution method at extract concentrations of 10%, 30%, 50%, 70%, and 90%. The MIC was determined based on the clarity of the medium after incubation, while the MBC was determined by subculturing the clear suspensions onto Mueller Hinton Agar and counting the bacterial colonies. The results showed that the broccoli ethanol extract contained flavonoids, alkaloids, saponins, and tannins. The MIC value against Staphylococcus epidermidis was found at a concentration of 50%. The average bacterial colony counts at concentrations of 50%, 70%, and 90% were 84 CFU, 13 CFU, and 3 CFU, respectively. The MBC value was not achieved within the tested concentration range because bacterial growth was still observed at the highest concentration tested. In conclusion, broccoli ethanol extract exhibited antibacterial activity against Staphylococcus epidermidis with a MIC value of 50%, while the MBC value was not obtained at concentrations up to 90%.
