Abstract

Salah satu Pemantapan Mutu Internal di Laboratorium yaitu dilakukannya pemeriksaan bahan kontrol, yang bertujuan untuk mengawasi kualitas hasil pemeriksaan. Penggunaan bahan kontrol menjadi sangat penting, namun dalam kondisi tertentu pengadaannya terbatas atau relatif mahal. Penelitian ini bertujuan untuk mengetahui stabilitas pooled sera yang ditambahkan propilen glikol sebagai bahan kontrol alternatif pada pemeriksaan kadar bilirubin total pada suhu refrigerator (6-8oC) dan suhu kamar (27-29oC). Pada penelitian ini, pemeriksaan sampel dilakukan setiap 3 hari sekali selama 30 hari terhadap 1 sampel secara triplo. Penelitian ini bersifat eksperimen. Pooled sera berasal dari kumpulan serum dengan kriteria tidak hemolisis, tidak ikterik, dan tidak lipemik. Tempat dan waktu penelitian dilakukan pada Februari 2020, di Laboratorium Kimia Klinik Jurusan Analis Poltekkes Bandung. Data yang dikumpulkan merupakan data primer yang diperoleh dengan memeriksa kadar bilirubin total metode DMSO (Dimetil Sulfoksida) pada pooled sera yang ditambahkan propilen glikol. Data dianalisis menggunakan statistika GLM (General Linear Model) metode repeated measures. Hasil penelitian menyimpulkan bahwa secara klinis dan statistik bilirubin total pada pooled sera yang ditambahkan propilen glikol pada suhu 6-8oC hanya stabil selama 3 hari dan pada suhu 27-29oC tidak mendapat kestabilan sampai hari ke-3. Secara klinis, pooled sera pada suhu 6-8oC stabil hingga 6 hari. Kata Kunci : Pooled Sera, Serum Kontrol, Stabilitas, Propilen Glikol, Bilirubin Total. Internal Quality Assurance in Laboratory is conduct examination of control materials, which try to check the quality of the examination results. The use of control materials is very important, but in certain conditions, the procurement is limited or relatively expensive. This study aims to determine the stability of pooled sera containing propylene glycol as an alternative control material in the examination of total bilirubin at refrigerator temperature (6-8oC) and room temperature (27-29oC). In this study, sample checking is once every 3 days for 30 days on 1 sample in triplo. This research is experimental. Pooled sera were collected by serums set with the criteria that not hemolysis, not jaundice, and not lipemic. The place and time of the study were conducted in February 2020, at the Clinical Chemistry Laboratory of Poltekkes Bandung. The data collected is the primary data obtained by examining the total bilirubin with DMSO (Dimethyl Sulfoxide) method in pooled sera containing propylene glycol. Data were analyzed using GLM (General Linear Model) statistics with repeated measures method. The results of the study concluded that clinically and statistically total bilirubin in pooled sera containing propylene glycol at 6-8oC was only stable for 3 days and at 27-29oC did not get stability until 3 days.
