Abstract

EFEKTIVITAS UREA TERHADAP PENGHAMBATAN SWARMING Proteus mirabilis TANPA MENGHAMBAT PERTUMBUHAN Staphylococcus aureus PADA AGAR DARAH Salma Nabilah Ridwansyah P17334117021 ABSTRAK Urea secara eksperimental telah terbukti memiliki sifat anti-swarming dan direkomendasikan untuk penggunaan laboratorium rutin. Ini umumnya digunakan dalam media kultur yang dirancang untuk identifikasi patogen infeksi saluran kemih termasuk Proteus spp. Tujuan penelitian ini untuk menentukan efektivitas urea terhadap penghambatan swarming P. mirabilis tanpa menghambat pertumbuhan S. aureus berdasarkan variasi konsentrasi urea 0.5%, 0.75%, 1%, dan kontrol 5%. Metode penelitian yang digunakan adalah eksperimental dengan menginokulasikan P. mirabilis dan S. aureus pada media agar darah berbagai variasi konsentrasi urea. Diameter zona swarming P. mirabilis yang terbentuk diukur dengan jangka sorong kemudian S. aureus diamati pertumbuhan koloninya. Hasil penelitian menunjukan semakin meningkat konsentrasi urea semakin menurun diameter zona swarming P. mirabilis serta untuk koloni S. aureus pada konsentrasi urea 0.5%, 0.75%, dan 1% terdapat pertumbuhan yang optimal. Hasil uji statistik Post Hoc Test Multiple Comparison dengan derajat kepercayaan 0.05 didapatkan nilai p value < 0.05 pada konsentrasi 1% dan kontrol 5% sehingga diketahui bahwa data pada konsentrasi tersebut memiliki perbedaan yang signifikan dengan konsentrasi lainnya. Dengan demikian dapat disimpulkan bahwa konsentrasi urea 1% merupakan konsentrasi yang efektif dalam menghambat swarming P. mirabilis tanpa menghambat pertumbuhan S. aureus. Kata Kunci : Swarming, Proteus mirabilis, Urea, dan Staphylococcus aureus EFFECTIVENESS OF UREA AGAINST INHIBITION OF Proteus mirabilis SWARMING WITHOUT INHIBITING THE GROWTH OF Staphylococcus aureus ON BLOOD AGAR Salma Nabilah Ridwansyah P17334117021 ABSTRACT Urea has been experimentally proven to have an anti-swarming properties and is recommended for routine laboratory use. This is commonly used in culture media designed to identify urinary tract infection (UTI) pathogens including Proteus spp. The purpose of this study was to determine the effectiveness of urea against the inhibition of P. mirabilis swarming without inhibiting the growth of S. aureus based on variations in the concentration of urea 0.5%, 0.75%, 1% and control 5%. The research method used was experimental by inoculating P. mirabilis and S. aureus on blood agar media with various variations of urea concentration. The diameter of the P. mirabilis swarming zone formed was measured by calipers and then S. aureus observed by its colony growth. The results showed that the increasing concentration of urea decreased the diameter of the P. mirabilis swarming zone and for S. aureus colonies at 0.5%, 0.75%, and 1% urea concentrations there was an optimal growth. The results of the Post Hoc Test Multiple Comparison statistical test with a degree of confidence of 0.05 obtained a p value < 0.05 at a concentration of 1% and a control of 5% so that it is known that the data at that concentration have significant differences with other concentrations. Thus it can be concluded that the concentration of 1% urea is an effective concentration in inhibiting the swarming of P. mirabilis without inhibiting the growth of S. aureus Keywords : Swarming, Proteus mirabilis, Urea, and Staphylococcus aureus
